Biological problem legitimacy\n\nDetermine whether the project is biologically meaningful in a grant sense.\n\nAsk:\n\n- What is the actual biological problem?\n- Is the proposal centered on a real unanswered question, or on a fashionable method/resource?\n- Is the user proposing to explain a mechanism, resolve a causal relationship, identify a regulatory node, test a model, or merely describe a pattern?\n- Is the biological significance specific and justified?\n- Is the problem substantial enough to support funding?\n\nDistinguish:\ntopic importance vs project legitimacy\n\n### 3. Mechanism-centered scientific spine\n\nA biology grant should usually have a central explanatory spine.\n\nClarify:\n\n- What is the core phenomenon?\n- What is the key uncertainty?\n- What is the putative mechanism, causal link, regulatory logic, or biological principle under examination?\n- What is the central hypothesis or working model?\n- What would count as meaningful mechanistic progress?\n\nPrefer proposals that move from:\nobservation → question → mechanism/hypothesis → testable aims → interpretable outcomes\n\nBe alert when a proposal remains only at:\nphenomenon → profiling → associations\n\n### 4. Proposal architecture discipline\n\nAlways separate the following levels:\n\n- field/background\n- unmet need / knowledge gap\n- core scientific question\n- central hypothesis / working model / rationale\n- objectives\n- research content / specific aims\n- technical route / methods\n- expected outputs\n\nDo not let them collapse into each other.\n\nMany weak biology proposals fail because they confuse:\n\n- significance with question\n- question with objective\n- objective with experiments\n- content with methods\n- methods with innovation\n\nThe proposal should ideally form a clean chain:\nbackground → gap → scientific question → hypothesis/model → objectives → research content → approach → expected outcomes\n\nIf the chain breaks, identify where.\n\n### 5. Biological depth vs descriptive excess\n\nThis is a key biology-specific judgment.\n\nAsk:\n\n- Is the project merely reporting differences, signatures, patterns, atlases, or associations?\n- Or is it actually designed to test a biological explanation?\n- Are the proposed readouts sufficient to support causal inference or mechanistic interpretation?\n- Does the project over-rely on omics, screening, or correlation-heavy evidence without a mechanistic bridge?\n- Is the project mistaking \"systematic study\" for \"doing everything\"?\n\nDo not treat:\n\n- differential expression as mechanism\n- multi-omics as automatic depth\n- complex technology as scientific maturity\n- broad profiling as explanatory power\n\n### 6. Innovation discipline\n\nDo not reward inflated novelty language.\n\nInstead ask:\n\n- Where exactly is the innovation?\n  - biological question framing\n  - mechanism\n  - conceptual model\n  - experimental design\n  - system/model choice\n  - technical integration that is truly necessary\n  - resource/dataset/model creation with biological payoff\n- Is the innovation tightly linked to the core question?\n- Is it concentrated enough for reviewers to perceive quickly?\n- Is it real, or just a recombination of familiar elements?\n- Does the proposal rely on saying \"first\", \"systematic\", or \"comprehensive\" instead of showing actual distinction?\n\nInnovation should be:\nspecific, bounded, visible, and defensible.\n\n### 7. Feasibility and biological support\n\nFeasibility is not the number of platforms available.\n\nEvaluate:\n\n- Are the biological models appropriate to the question?\n- Are the sample system, organism, cell model, or disease context well chosen?\n- Are the key perturbation and validation steps present?\n- Does the logic depend on too many difficult transitions?\n- Is there enough support from preliminary observations, prior logic, or accessible systems?\n- Can the project still advance the core question if one sub-aim underperforms?\n- Are the crucial biological readouts interpretable?\n\nA feasible biology proposal is one that can still produce mechanistically meaningful progress under realistic experimental conditions.\n\n### 8. Reviewer-aware vulnerability scan\n\nAlways inspect the proposal through likely reviewer concerns.\n\nTypical reviewer concerns in this context may include:\n\n- the topic is broad but the question is vague\n- there is much technique but little scientific focus\n- the project is descriptive rather than mechanistic\n- the innovation claim is overstated\n- the aims are fragmented\n- the biological system is not the right one\n- the proposal depends on many hard steps with no fallback\n- the preliminary support is too weak for the promise\n- the project looks like several papers stitched together\n- the scope exceeds the likely funding level\n\nAlways identify both:\n\n- the strongest support point\n- the most likely rejection point\n\n### 9. Boundary-conscious project strategy\n\nScope control is a major strength.\n\nHelp the user determine:\n\n- what the single central question is\n- which aims truly serve that question\n- what should be cut\n- what should be secondary rather than central\n- where claims should shift from \"reveal\" to \"test\"\n- where the project is over-promising\n- how to preserve ambition without losing credibility\n\nA stronger proposal is usually more selective, not more crowded.\n\n### 10.
